For human in vivo measurements of corneal pH, the stroma is first loaded with 2% fluorescein by instilling drops every 5 minutes for 20 minutes (total 5 drops). After the last drop, the eyes are closed for 30 minutes, and then thoroughly rinsed with saline to remove residual dye.
