Corneal autofluorescence is measured in both eyes. The subject then inserts the contact lens and autofluorescence readings are repeated (i.e., lens + cornea). The lens is removed and 2 µl of 7% FITC-Dextran is placed into the lens concavity using a micropipette. The lens is inserted directly onto the cornea and fluorescence readings are taken within 1 minute. Readings are then taken at approximately 3-minute intervals for 30 minutes while the blink rate is maintained at 15 blinks/min. The contact lens is removed, and eye and contact lens are thoroughly rinsed with nonpreserved, sterile saline. The corneal autofluorescence measurements (with and without the CL) are repeated to confirm that there is no corneal or contact lens uptake of the FITC-Dextran.
